为了使取出的样品更具代表性,体细胞应均匀分布于观察腔内。文中利用微流控芯片代替传统的载玻片作为观察牛乳体细胞的平台。体细胞由于传统载玻片的边缘效应会聚集在载玻片中央,导致细胞粘附。使用载玻片对操作人员的要求很高,当覆盖层被盖玻片覆盖时,由于处理不当可能会引入气泡。微流控芯片则可以利用压力泵将溶液直接注入芯片内部,并在芯片内部产生层流,没有边缘效应也不会产生气泡。此外,注射操作简单无需培训即可完成。
为提高计数精度,取九张图片计数结果的平均值作为最终的体细胞数量。为保证在一个观测腔内拍摄九张不同区域的图像,观测腔面积必须较大。根据PDMS的特性,如果观测腔面积过大,则会导致观测腔中心塌陷,所以观测腔面积过大无法保证体细胞分布的均匀性。因此,提出了九宫格型的微流控芯片结构[9]。在每个观测腔上方直接拍摄一张图像,以确保体细胞的均匀分布,同时确保拍摄区域不重复。
通过仿真粒子在观测腔内的分布状况来验证牛乳体细胞在微流控芯片观测腔内的分布状况。为了使粒子在微流控芯片的九个观测腔内分布最为均匀,使用Comsol仿真观测腔直径与流道宽度在不同比例下粒子的分布情况,仿真过程中使观测腔的半径固定,变化流道宽度,选用流体流动颗粒跟踪物理场和层流物理场进行耦合。层流场的流体材料选择和生鲜牛乳具有同种密度和动力粘度的材料替代。密度取1039 $ \mathrm{k}\mathrm{g}/{\mathrm{m}}^{3} $,动力粘度取0.0017 Pa·s。流体进入芯片入口的速度为0.02 m/s,以此模拟生鲜牛乳的进样速度。因为牛乳体细胞中白细胞占比为98%,因此释放的粒子选择和白细胞具有同直径的粒子替代,直径取14 μm,每0.05 s释放一次粒子,共释放1 s的时间,粒子的初始速度取决于流体的速度场。粒子的运动行为遵循牛顿第二定律。
观测腔半径与流道宽度在不同比例下的粒子分布,如图1(a)所示。通过图1(a)发现,若流道宽度过窄会使粒子集中在观测腔的中间位置,不会均匀地充满整个观测腔;若流道宽度过宽又会使粒子大量堆积在观测腔内,造成多个粒子重叠的现象。综合流体压力、流体速度等对通道的影响,最终确定观测腔的半径与流体通道的宽度比例为25:7,粒子刚好可以均匀地充满整个观测腔。通过Comsol计算得出的九宫格型微流控芯片的粒子分布仿真图如图1(b)所示。通过仿真图可以直观地看出,粒子在微流控芯片的九个观测腔内分布比较均匀,因此,九宫格型微流控芯片可以满足体细胞分布均匀性的要求。

芯片的制备过程可简单描述为:首先,采用光刻技术[10]制造出九单元微通道凸突的阳模,如图2(a)所示。光刻步骤如图2(b)所示。其次,在阳模上浇铸聚二甲基硅氧烷(Polydimethylsiloxane, PDMS),PDMS在一定温度下固化后便可从阳模上剥离,制成带有微通道的芯片基片。最后,用玻璃盖片封接后制成高分子聚合物微流控芯片。其过程如图2(c)所示。在九宫格微流体网络上设置一组用于装载被测样本的微流体微腔,微流体流路的一端连接到入口,另一端通过出口与废液腔连接。在设计中,微流体通路上平均分布着三个观测腔,整个微流控芯片共有三个通路,九个观测腔,观测腔半径为500 μm。显微成像系统采集观测腔处的细胞图像信息。为了减少细胞在观测微腔中的聚集以及在进样通道中的贴壁情况,使细胞顺利地在进样通道流动,设计微流体的通道高度为40 μm,通道宽度为140 μm。九宫格微流控芯片示意图如图2(d)所示。

细胞的计数算法分为图像处理和细胞计数两个部分,图像处理的目的是为了改善图像的识别效果,突出目标信息,降低背景信息对目标信息的影响,使处理后的图像更利于细胞计数。图像处理的过程分为图像灰度化、图像滤波、图像增强、Canny边缘算子检测和形态学开运算。细胞计数是将经过图像处理后的图像通过连通域计数得到体细胞数量。
牛乳体细胞通过瑞氏染色法[11]染色后,细胞核显示蓝紫色颗粒状,其他区域呈浅粉色。原始图像如图3(a)所示(为更清楚地说明图像处理细节,以拍摄图像的1/9为例)。通过显微成像系统拍摄出来的原始照片为彩色图像,也即RGB图像。灰度图像占用的计算机内存小,运算速度快,便于后续处理,因此,先进行图像的灰度化处理。

选用加权值法[12]来进行图片的灰度处理,如公式(1)所示:
$$ Gray=0.114Blue+0.587Green+0.299Red $$ (1) 式中:$ Red $的权重取0.299;$ Green $的权重取0.587;$ Blue $的权重取0.114,通过加权值法转换出的灰度图像,既可以将背景与目标很好地区分出来,又可以提升图片的亮度,同时又不会增加图片的噪声。灰度处理后的图像如图3(b)所示。
在拍摄、存储、传输照片的过程中,由于成像系统或者是传输介质等原因,不可避免地使图像受到噪声的污染。因此,有必要滤除这些杂质以提高图像质量。图像通常含有脉冲噪声或高斯噪声。利用中值滤波[13]可以抑制脉冲噪声,中值滤波如公式(2)所示:
$$ g(i, j)= \mathop{Med } \limits_A \{f(i, j)\} $$ (2) 式中:$ g\left(i,j\right) $为中值滤波的输出结果;$ Med $表示取中值操作;$ f\left(i,j\right) $为输入序列;A为邻域窗口,窗口尺寸取奇数,文中选用3×3。高斯滤波可以很好地滤除掉图像中的随机高斯噪声。但是高斯滤波在滤波的过程中只关注了位置信息,会过滤掉图片的边缘和纹理信息,所以在进行高斯滤波后,图像会变得模糊。而双边滤波在高斯滤波的基础上增加了像素值权重项,即在考虑距离因素影响的同时也要同时考虑由于像素值差异所带来的影响,像素值越近,权重越大。使用双边滤波[14]可以在保证图像边缘和纹理信息不被破坏的情况下滤除高斯噪声,双边滤波如公式(3)所示:

对灰度图像进行滤波时,在过滤掉噪声的同时会造成轻微的图像模糊。为去除图像模糊,首先通过矩阵的掩膜操作[15],增加了图像的对比度,使图像变得更清晰。然后通过对比度受限制自适应直方图均衡法(CLAHE)[16],进一步提高图片的亮度和对比度。CLAHE通过限制局部直方图的高度来限制噪声放大和局部对比度增强。CLAHE会选取一个滑动的模板,模板的映射函数$ n $(i)如公式(5)所示:
$$ n\left(i\right)=\frac{255\times {C}_{DF}\left(i\right)}{N\times N} $$ (5) 式中:N×N为滑动模板的大小; $ {C}_{DF}\left(i\right) $为原始图像的累积直方图。模板划到的区域内直方图的累计分布函数如公式(6)所示:
$$ S=\frac{{\rm{d}}\left(n\right(i\left)\right)}{{\rm{d}}i}={H}_{ist}\left(i\right)\times \frac{255}{N\times N} $$ (6) 从上式可以看出,通过控制灰度直方图的高度可以控制映射函数的斜率,以此来限制对比度的强度。直方图的最大高度$ {H}_{{\rm{max}}} $如公式(7)所示:
$$ {H}_{{\rm{max}}}={k}_{{\rm{max}}}\times \frac{255}{N\times N} $$ (7) 式中:$ {k}_{{\rm{max}}} $为斜率的最大值,对于高度大于直方图最大高度$ {H}_{{\rm{max}}} $的直方图,应当截去多余的部分。在CLAHE的实际应用中,可以设定一个截断阈值,截断阈值可以不等于灰度直方图的最大高度,使截断的部分在整体的灰度范围内分布均匀,可以在直方图总面积不变的前提下提升直方图的高度。增强后的图像如图3(d)所示。
在对牛乳体细胞进行计数时,不关心体细胞内的具体信息,只需要提取体细胞外部轮廓即可。文中采用Canny边缘检测算法[17]对经过图像增强后的牛乳体细胞进行边缘提取,其算法流程图如图4所示。经过Canny边缘算子检测后输出的图像为边缘的二值图像,如图3(e)所示。灰度图像中残余的一些噪声点也会显露出来,此时可通过形态学操作中的开运算[18]将这些微小的噪声点去除。开运算是对图像先腐蚀后膨胀的操作,用来去除小物体、平滑大物体边界且不明显改变原来区域的面积。通常在需要去除小颗粒噪点以及分离纤细点处目标之间的粘连时使用。与腐蚀操作相比,其优势在于能基本保持目标原有的大小不变。最终输出图像如图3(f)所示。其定义如公式(8)所示:
$$A \circ B=(A \Theta B) \oplus B $$ (8) 图像的连通域[19]是指图像中具有相同像素值并且由位置相邻的像素组成的区域。图像中每个细胞的边缘组成了一个连通域,通过计算图像中连通域的个数,就可以得到图像中的体细胞个数。

在当地的牧场随机选取了20头奶牛通过上文所述的牛乳体细胞计数系统对其分泌的牛乳在现场进行体细胞个数的检测实验,以验证体细胞分布的均匀性和检测乳房的健康状况。每头奶牛均是在挤奶完成后的1 h内完成对体细胞的计数,以保正牛乳的生鲜性。

为了使取出样品中的体细胞能够反应奶牛的乳房健康状态,体细胞应均匀分布于观察腔内。为此随机抽取一头奶牛将其牛乳泵入九宫格型微流控芯片,使用显微成像系统对其微腔进行拍摄,基于九宫格型微流控芯片拍摄出的牛乳体细胞图像如图7(a)所示。使用相同的牛乳通过载玻片制作牛乳体细胞切片,使用显微成像系统对其进行拍摄,基于载玻片拍摄的牛乳体细胞图像如图7(b)所示。通过图7可以直观地看出,若使用载波片则无法保证牛乳体细胞分布的均匀性,牛乳体细胞大量聚集在载玻片的中心位置,造成大量细胞黏连,形成细胞团,体细胞分布不均匀,通过这种方法计算出整体的体细胞数量与其真实值之间有较大的误差,导致对奶牛乳房的健康状况做出错误的判断。而基于九宫格型微流控芯片拍摄出的体细胞分布均匀,与仿真结果一致。

对基于九宫格型微流控芯片细胞分布均匀性做进一步分析,将20头奶牛分别赋予1~20的编号,对每号奶牛牛乳拍摄9张图像,为表述清楚,将第1号奶牛牛乳的9张图像命名为第1组图像,第2号奶牛牛乳的9张图像命名为第2组图像,依次类推。每张图像牛乳体细胞数量如表1所示,以一组图像为例,图像的实拍图如图8所示。计算各组图像体细胞标准差系数,如表1倒数第2列所示。由于每组图像体细胞数量的平均值不同,对于不同水平的总体不宜直接用标准差指标进行对比,因此使用标准差系数来衡量每一组图像中体细胞个数的离散程度,即离散程度越小,体细胞分布也越均匀。由表1数据可知,每组图像标准差系数的最大值为1.61%,最小值为0.21%,平均值为0.69%,认为芯片9个观测腔内的体细胞数量波动在可允许范围内,证明体细胞在微流控芯片的9个观测腔内分布均匀。

综上,相较于载玻片,基于九宫格型微流控芯片极大地提高了牛乳体细胞分布的均匀性,避免了样品因为细胞粘附性、重力等原因造成的细胞沉降、贴壁等问题,从而使体细胞在样品溶液分布均匀,保证了取出的样品具有代表性,提高了奶牛乳房健康状态的判断准确率。

取每组9张原始照片自动计数的平均值作为自动测量值,将每组9张原始图像中手动计数的平均值作为手动测量值。根据图像参数,图像长度5802 μm,宽度为4050 μm,通道高度为40 μm,视场内溶液体积如公式(9)所示:
$$ \begin{split} V= & 5 \; 802 \times 4 \; 050 \times 40=9.399 \; 24 \times 10^8 \; {\text{μ}} \mathrm{m}^3= \\& 0.94 \; \mathrm{~mm}^3=0.94 \; {\text{μ}} \mathrm{L} \end{split} $$ (9) 由公式(9)可得视野范围内牛乳的体积为0.94 μL。每毫升生牛乳中体细胞数可以从每组图像体细胞数的平均值得到。BD FACSAriaTM流式细胞仪计数作为真值,计算手动计数和自动计数的准确性。每毫升生牛乳体细胞数的手动测量值、自动测量值和真实值的比较如图9(a)所示。可以看出,手动测量值和自动测量值都很接近真值。测量值与真实值之间的误差分布如图9(b)所示。

从图9(b)可以看出,自动计数的相对误差绝对值的最大值为2.93%,最小值为0.53%,平均值为1.72%,得出自动计数的最大计数误差为±2.93%。手动计数相对误差绝对值的最大值为1.18% ,最小值为0.24% ,平均值为0.63%,手动计数的最大计数误差为±1.18%。可以认为,自动计数相对误差和手动计数相对误差较小,均在允许范围内。自动计数的准确率在97.07%~99.47%之间,手动计数的准确率在98.82%~99.76%之间。
该系统自动计数的准确率比手动计数的准确率略低,主要原因是牛乳体细胞中包含多种不同的细胞,其中淋巴细胞的直径在5~12 μm之间,中性粒细胞的直径在9~12 μm之间,乳腺上皮细胞的直径在12~18 μm之间,巨噬细胞的直径在15~25 μm之间,四种体细胞如图10所示。当两个较小体细胞距离较近或发生重叠时,系统自动识别能力较差,不能分辨出是两个不同的小体细胞还是一个较大的体细胞,因此导致体细胞计数产生误差。由于基于九宫格型微流控芯片使体细胞分布的均匀性高,很少出现上述情况,所以自动计数和手动计数都有很高的准确率。自动计数相较于手动计数更加便捷、省时。自动计数无需人员操作由系统自动完成,缩短了检测时间使整个检测过程可控制在1 min内;手动计需要操作人员进行操作,检测时间依赖于操作人员的熟练程度,没有自动计数便捷。

在不同的研究中,奶牛牛乳中体细胞数对奶牛乳房健康的判断标准不尽相同[23-27]。结合多种因素,文中推导出判断奶牛乳房健康状况和疾病程度的标准,如表2所示。测量值和真实值的判断结果如表3所示。

当挤牛乳时,怀疑患有亚临床乳腺炎的奶牛应该在牛群的最后挤奶。如果条件允许,可以为它们配备单独的挤奶器,这样就不会干扰其他健康的奶牛。已经患有亚临床乳腺炎的奶牛必须被隔离和治疗,以避免牧场的进一步损失。
综上所述,建议的挤奶顺序为20号、6号、9号、2号、12号、4号、1号、18号、3号、14号、5号、10号、15号、17号。建议的治疗顺序为11号、13号、16号、7号、19号、8号。
A study on the uniform distribution and counting method of raw cow's milk somatic cells based on microfluidic chip

摘要: 生鲜牛乳中的体细胞数量是判断奶牛是否患有乳房炎的重要依据。针对牛乳在取样过程中细胞贴壁沉降等原因造成体细胞分布不均匀,从而导致体细胞计数不具有代表性的问题,文中提出了一种基于九宫格型微流控芯片使体细胞分布均匀并提升计数准确率的方法。首先在Comsol仿真的基础上制备了九宫格型微流控芯片,提高了细胞分布的均匀度。其次研制了集染色、搅拌于一体的负压进样系统,保证在进样过程中持续保持体细胞分布的均匀度和不受空气的污染。并配合芯片研制了微型显微成像系统,对芯片的九个观测腔拍摄图像。最后通过图像处理的方法对体细胞进行计数,并判断奶牛乳房的健康状况。实验结果表明,每组九张图像体细胞数量的标准差系数均小于等于1.61%,系统计数准确率可达到99.23%。该研究方法为奶牛乳房炎的检测与预防奠定了基础。Abstract:
Objective The somatic cell count (SCC) in raw milk is an important basis for determining whether a cow is suffering from mastitis. Identifying cows with mastitis by testing the SCC, then isolating and treating them as early as possible, can effectively prevent the spread of bacteria in the herd to reduce consequential economic losses. However, traditional methods may lead to uneven distribution of somatic cells during milk sampling, such as cell adhesion settlement, and unrepresentative somatic cell count due to lack of matching imaging system. In this paper, a method is proposed which is based on the nine-cell grid microfluidic chip to make somatic cell evenly distributed and develop a two degree of freedom displacement platform equipped with a micro lens to improve the counting accuracy . Methods Firstly, a simulation was performed to verify the uniformity of the somatic cell distribution within the chip observation cavities (Fig.1). And based on the simulation results, a nine palace grid microfluidic chip was prepared (Fig.2). Secondly, a two-degree-of-freedom displacement platform (Fig.6) equipped with a micro-camera lens is developed, which can automatically take images of the nine observation cavities of the chip, making image acquisition more convenient. Finally, somatic cells were counted by image processing (Fig.3), so as to verify the uniformity of somatic cell distribution, obtain the counting accuracy, and judge the health status of cow udder. Results and Discussions 20 cows were randomly selected from local pastures to verify the performance of the proposed method. From the data in Tab.1, it can be seen that the standard deviation coefficient of the SCC in each group of nine images is less than or equal to 1.61%, which verifies the uniformity of the distribution of somatic cells in the nine observation cavities of the microfluidic chip. The system ensures the uniform distribution of somatic cells and renders the taken samples more representative. As can be seen from Fig.9(b), the maximum absolute value of the relative error of the system of automatic counting is 2.93%, the minimum value is 0.53%, and the average value is 1.72%. The maximum relative count error of the automatic counting is obtained as ±2.93%. The system has a very high accuracy for automatic counting. Conclusions The experimental results show that the somatic cell counting system developed in this paper can make the somatic cell distribution in fresh milk more uniform and count more accurately. The standard deviation coefficient of the number of somatic cells in each group of nine images was less than or equal to 1.61%, and the smaller the standard deviation coefficient is, the more uniform the distribution of somatic cells is. The accuracy of the automatic system counts ranged between 97.07% and 99.47%. This research method lays the foundation for the detection and prevention of mastitis in cows. -
